چکیده

ABSTRACT Soybean crops grown in the plains of eastern Colombia have been affected by incidence rust. This disease, still officially regarded as American soybean rust (caused Phakopsora meibomiae) Colombian Department Agriculture (ICA), causes serious damage to growing areas. Symptoms rust, such reddish-brown lesions, observed since 2004 upper half plants during vegetative stages. In 2005 and 2018, infected leaf tissues uredinospores were collected from an experimental area commercial fields. Once morphological identification Koch’s postulates confirmed presence spp., molecular characterization was performed identify pathogen associated with disease species level. based on standard procedures using internal transcribed spacer (ITS) regions rDNA PCR amplification specific primers for meibomiae P. pachyrhizi. The obtained sequences BLASTed against GenBank/NCBI data bank. Results indicated that pachyrhizi Sydow is fact causative agent Colombia, considering samples 2018. ITS-rDNA deposited at GenBank under accession numbers MK933723 MK933731. finding reported ICA, so they could update phytosanitary status this Colombia.

Expressed sequence tag analysis of the soybean rust pathogen Phakopsora pachyrhizi.

Soybean rust is caused by the obligate fungal pathogen Phakopsora pachyrhizi Sydow. A unidirectional cDNA library was constructed using mRNA isolated from germinating P. pachyrhizi urediniospores to identify genes expressed at this physiological stage. Single pass sequence analysis of 908 clones revealed 488 unique expressed sequence tags (ESTs, unigenes) of which 107 appeared as multiple copie...

From Select Agent to an Established Pathogen: The Response to Phakopsora pachyrhizi (Soybean Rust) in North America.

The pathogen causing soybean rust, Phakopsora pachyrhizi, was first described in Japan in 1902. The disease was important in the Eastern Hemisphere for many decades before the fungus was reported in Hawaii in 1994, which was followed by reports from countries in Africa and South America. In 2004, P. pachyrhizi was confirmed in Louisiana, making it the first report in the continental United Stat...

Correlations between traits in soybean (Glycine max L.) naturally infected with Asian rust (Phakopsora pachyrhizi).

Soybean (Glycine max L.)-breeding programs aim to develop cultivars with high grain yields and high tolerance to Asian soybean rust (Phakopsora pachyrhizi). Considering that the traits targeted for breeding are mainly quantitative in nature, knowledge of associations between traits allows the breeder to formulate indirect selection strategies. In this study, we investigated phenotypic, genotypi...

Development of simple sequence repeat markers for the soybean rust fungus, Phakopsora pachyrhizi.

Twenty-four simple sequence repeat markers were developed for Phakopsora pachyrhizi, a fungal pathogen of soybean (Glycine max) and other legumes. All 24 of the loci were evaluated on 28 isolates of P. pachyrhizi. Twenty-one loci were polymorphic, with allelic diversity ranging from two to eight alleles, and null alleles were observed for eight of the 24 loci. A preliminary screen with the clos...

A Small Cysteine-Rich Protein from the Asian Soybean Rust Fungus, Phakopsora pachyrhizi, Suppresses Plant Immunity

The Asian soybean rust fungus, Phakopsora pachyrhizi, is an obligate biotrophic pathogen causing severe soybean disease epidemics. Molecular mechanisms by which P. pachyrhizi and other rust fungi interact with their host plants are poorly understood. The genomes of all rust fungi encode many small, secreted cysteine-rich proteins (SSCRP). While these proteins are thought to function within the ...
